hfCas12Max® 是对 xCas12i DNA 编辑工具进行蛋白工程化改造、筛选及验证,开发出的高保真变体,相较传统的 Cas9 和 Cas12,hfCas12Max® 具有更宽泛的 PAM 识别和更高效的编辑效率,且相当或更低的脱靶效应。hfCas12Max®识别 5'-TN 或 5'-TNN PAM 序列,利用其单一 RuvC 结构域实现对 dsDNA 的切割,形成 DNA 双链断裂(DSBs),利用细胞的 DNA 修复机制实现基因的敲除与敲入。
